Interferon-y Upregulates Interleukin-3 (IL-3) Receptor Expression in Human Endothelial Cells and Synergizes With IL-3 in Stimulating Major Histocompatibility Complex Class I1 Expression and Cytokine Production

چکیده
The human interleukin-3 (IL-3) receptor is constitutively expressed on certain hematopoietic cells where it mediates proliferation and differentiation, or functional activation. We have recently found that human umbilical vein endothelial cells (HUVECs) also express IL-3 receptors and that the expression is enhanced by stimulation with the monokine tumor necrosis factor a. In this report we show that the lymphokine interferon y (IFNy) is a powerful stimulator of the IL-3 receptor of HUVECs and that the combination of IL-3 and IFNy has a synergistic effect on major histocompatibility complex (MHCI class II expression and on the production of the early-acting hematopoietic cytokines IL-6 and granulocyte colony-stimulating factor (G-CSF). IFNy caused a timeand dose-dependent up-regulation of mRNA for both the LY and j3 chnins of the IL-3 receptor, with maximal effects occuring 12 to 24 hours after stimulation with IFNy at 100 U/ mL. Induction of mRNA correlated with protein expression on the cell surface, as judged by monoclonal antibody staining of both receptor chains and by the ability of HUVEC to specifically bind '251-labefed IL-3 ['a51-lL-3). Interleukin 10(EL,.10) Inhibits Cytokine Synthesis by Human Monocytes: An Autoregulatory Role of IL-10 Produced by Monocytes By Ren6 de Waal Malefyt,
In the present study we demonstrate that human monocytes activated by lipopolysaccharides (LPS) were able to produce high levels of interleukin 10 (I1-10), previously designated cytokine synthesis inhibitory factor (CSIF), in a dose dependent fashion. I1-10 was detectable 7 h after activation of the monocytes and maximal levels of I1-10 production were observed after 24--48 h. These kinetics in...
متن کاملThe Role of Interleukin (IL-22) in immune response to human diseases
Background and aims: IL-22 is an alpha- helical cytokine. IL-22 binds to a heterodimeric cell surface receptor composed of IL-10R2 and IL-22R1subunits. IL-22R is expressed on tissue cells, and it is absent on immune cells. L-22 and IL-10 receptor chains play a role in cellular targeting and signal transduction to selectively initiate and regulate immune responses. The aim of this study was to i...

OBJECTIVES To investigate the influence of the Th1 and Th2 lymphokines interleukins (IL)-4 and IL-13, interferon gamma (IFN gamma), and several monokines on the adhesion of mononuclear cells to synovial fibroblasts and intercellular adhesion molecule-1 (ICAM-1) expression and cytokine production of synovial fibroblasts in patients with osteoarthritis. METHODS Synovial fibroblasts were isolate...
متن کاملEffects of histamine and its antagonists on murine T-cells and bone marrow-derived dendritic cells
We determined the effects of histamine and its antagonists on the surface marker expression of dendritic cells (DCs) and the influence of lipopolysaccharide (LPS), histamine, and histamine receptor antagonists on DCs and T-cells. The bone marrow was extracted from the femurs and tibiae of 6- to 8-week-old female Balb/c mice and cultured in medium containing penicillin, streptomycin, L-glutamine...
متن کاملThe Effect of Beta Interferon on Dendritic Cells and Cytokine Synthesis by CD4+ T Cells
Background: Dendritic cells (DC) are a key regulator of the immune response, and interferon- beta (IFN-β) is considered an immunomodulatory molecule for DC. Objective: The purpose of this study was to evaluate the ability of IFN-β treated DC to induce cytokine secretion by CD4+ T cells. Methods: Dendritic cells were generated from blood monocytes with granulocyte-monocyte colony-stimulating fac...
